Protein Expression Profiling: Current Trends and Future Horizons

Protein expression and quantitation is attracting considerable interest
in the life science community and is developing as one of the major
challenges in defining mechanisms of action in biological systems. Mass
spectrometry, in its variety of forms, holds significant promise for
determining relative levels of protein and peptides in complex mixtures.
In this short presentation, chemical and instrumental methods will be
discussed that allow quantitative analysis of peptides from complex
mixtures. Reagents like ICAT and TMPP (2,4,6-trimethoxy phenol
phosphonium acetic acid N-hydroxysuccinimide ester) will be compared and
contrasted. Additionally, appropriate analytical methods and software
that allow peptides to be analyzed with greater specificity and in some
cases greater sensitivity will be discussed. Software that then allows
for the extension of peptide quantitative measurements to protein
expression will also be presented.
