Rapid Separations Using Eksigent's NanoLC Platform

By combining the core technologies of microfluidic flow control, microscale pumping, and microfabrication, Eksigent is creating entirely new classes of analytical instruments and fluid delivery systems for drug discovery applications. These systems create levels of throughput, information content,and miniaturization not previously possible. Eksigent's products include the NanoLC system for high throughput chromatography and proteomics research and the EKPump for precise, microscale fluid delivery in medical devices.

The Eksigent NanoLC System delivers HPLC performance in a capillary format. Optimized for 300 µm columns at flow rates of 200 nL/min to 10 µL/min, the NanoLC System includes UV absorbance detection using a microfabricated flow cell with integral fiber optics. The capillary column format reduces solvent and sample use by more than 99% compared with 4.6 mm columns. This system uses a microfluidic flow controller (MFC) to provide rapid, precise adjustment of a variable pressure source. With MFC technology, true microscale chromatography is achieved with direct pumping: flow splitting is not required. Flow meters actively measure the actual flow rate in both mobile phases, and the electronic MFC controller provides immediate adjustments to deliver an accurate, reproducible gradient run after run.

Since the flow controller is miniaturized, it can be easily configured as a multiplexed high throughput system. The NanoLC-8X is ideal for high-throughput HPLC for ADME/Tox and method development. This system can also be configured for multidimensional chromatography for automatic, parallel 2D chromatography interfaced to MALDI targets.
