Gilson Dimensions LC System:
High throughput 2-D and 3-D separations Via Automated SPE

Multi-dimensional separations (commonly 2-D or 3-D) are useful to selectively isolate samples for LC-MS/MS analysis and to minimize ion suppression. A 2-D LC separation typically involves an ion-exchange, affinity, size exclusion or chromatofocusing column coupled with a reversed phase column in either the first or second dimension. 2-D gels are also used for dual separations; here, two-dimensional electrophoresis separates proteins in the first dimension by isoelectric focusing based on isoelectric point and in the second dimension by molecular weight using sodium dodecylsulfate polyacrylamide gel electrophoresis. Since these approaches have some limitations, an alternative is the Gilson Dimensions System which develops high throughput 2-D and 3-D separations via automated solid-phase extraction (SPE).

Multidimensional separations via SPE are performed using disposable cartridges packed with sorbent phases (100mg/1mL). The liquid handler (modified Model 215) conditions the cartridge, loads, washes and elutes multiple sample fractions, as shown below.




Advantages of Multidimensional Separations via SPE:

  • No complicated plumbing or timing of valve switches; the only plumbing required is for the Injection valve
  • Fast fraction preparation time (<1 hr for 30 fractions)
  • Removes the risk of carry over from column since cartridges are used only once
  • Increased flexibility; can add additional dimensions without significantly increasing prep time, plumbing complexity, or system cost
  • An economical system (cost is equivalent to a standard HPLC system)
  • Completely automated
