Vendor Session - Liquid Chromatography-based Assays for Secondary Screening

Steve Hobbs, Nanostream

  • Veloce µPLC system
    • Kinase assay
      • Separation Conditions: C18, 7 m; 30 mm x equiv. i.d. 0.5 mm; 600 mL/min (25 mL/min/col).
    • Advantages of µPLC vs. plate reader
      • Separation before detection reduces interferences by separating quenchers and fluorescent compounds from substrate
      • Fluorescent quenching groups are not necessary; labels do not need to be placed close to enzyme’s activation site
      • Veloce system offers predictability in assay development time, with reproducible results
